43 tooth sprocket transformed my TT250 into a usable motorcycle. I was into 5th gear at 35mph with the 50 tooth rear sprocket. I think mine was shipped with the wrong primary gearing (between the crankshaft and gearbox) I have nothing to prove that but my experience with old motorcycles and gearing.
