If you seal it off the motor will die from fuel starvation. There will be no way for the air to replace the consumed fuel and therefore the fuel will lock itself in the fuel tank causing the motor to die.
Some newer cars as well as off road jeeps come with a check valve in the fuel cap to prevent fuel spillage in the event of a roll over. I'm sure you could rig something similar.
