250 transmissions clicking in first

So basically I have a shark 250, basically a hawk 250 with rear drum brakes and counterbalanced, if theirs a clicking in first gear does that mean I need to replace the gearbox? And if so where can I get the part and how is it replaced? Do I need to remove the engine? Is there a YouTube video?

Watch as many tranny-repair Youtube vids as you can find.
Take lots of pics as you disassemble
Try to find a good m/c tranny repair manual

I'm sure you've checked the condition of the sprockets and chain, as well as the tension. A stiff link on a chain can make some noise.
