So I replaced my back tire and went for a ride. Caliper fell off :( I didn't apply new loctite. Do I need to reapply every time?

Every time you remove a bolt/nut it will need more 'Locktite/Threadlocker' applied to the threads

The rear caliper fell off my friend's brand new Harley on his first out of state trip. It didn't end well when the rear wheel locked up on a high speed corner. Insurance company totaled the bike and he bought a new Harley. Recurring EFI problems with that one but that's another story.
